Effect of Taraxacum officinale extract against gentamycin‐induced reproductive damage in male rats.

lntroduction: Gentamicin (G) is an aminoglycoslde antibiotic that has a broad spec..

trum agalnst microbial pathogens, especially gram-negative bacteria. However, it is well

known that gentamicin has some adverse effects such ototoxicity and testicular toxicity.

Taraxacum officinale extract (TOE) has been widely used as a folkloric medicine for the

treatment of diverse diseases. Furthermore, TOE has inhibiting effect on reactive oxygen

species. The aim of this study was to investigation of the effects of TOE on gentamycin

induced reproductive damage in male rats.

Material and methods: in the study, 24 male Sprague Dawley rats aged eight weeks

old and weighted 250-300 gr, were used in this study. The animals were obtained from

Atatürk University Experimental Research Centre and housed in standard laboratory conditions.

Commercial pellet chow and fresh drinking water were available ad libitum. Rats

were divided into four groups. 1. group (n= 6) referred as control group and physiological

saline via oral gavage to the animals il. group (n=6) referred as Gentamicin (G) and

2-mg;kg gentamicin sulphate (GS) was injected intraperitonally (iP) to animals. 111. Group

(n=6) was referred as G + TOE150, 2-mglkg GS and 150 mglkg TOE injected iP to animals.

iV. group was referred as G+ TOE200 (n=6), 2-mglkg GS and 200 mg/kg TOE injected

iP to anlmals. The treatment was performed fer consecutive 7 days. The animals were

tranquilized and sacriflced on sııı day of study. Cauda epididymal semen samples and

testes tissues were collected. Routine semen examinations were performed and oxidative

stress levels of testicular tissues were assayed. Reproductive organ weights (total testes

weight (TlW) and total cauda epididymides weights (TCEW) ] were recorded. Velocity

of sperm cells in G, G+TOE150 and G+TOE200 groups were signiflcantly lower when

compared to control group. On the other hand, motility in G group was significantly lower

when compared to control group. However, !here were no difference between in treatments

G+TOE150, G+TOE200 groups. Furhermore, there were ne differences in terms

of dead sperm rate, sperm density, total abnormality, TIW and TCEW among ali groups.

MDA level of G group was significantly higher than in control and treatment groups.

Glutathione peroxidase (GPx) and glutathione (GSH) levets in G group were lower than

the other groups. The lowest SOD level was in G group while the highest level was in

control group. MDA level was higher and CAT level was lower in G group when compared

to other groups.

Conclusion: The dose of G +150mg/kg and G +200 mg/kg TOE is effective on mentioned

spermatological parameters induced by gentamicin.
