Kitosan-Gümüş Nanopartikül Yüklenmiş Oleuropein'in Ratlarda Streptozosin ile İndüklenen Diyabet Üzerindeki Etkilerinin Araştırılması

Abstract:

Diabetes is a metabolic disease characterized by high blood glucose levels and is widely prevalent across the world. It has adverse effects on various organs, including the heart, kidneys, digestive system, and nervous system. Chitosan-Silver Nanoparticles (K-AgNP) offer significant advantages for drug delivery systems; they enhance the stability and solubility of components, improve intermembrane transport capability, and prolong the drug distribution period. Oleuropein, the most prominent phenolic compound in olive leaves, exhibits numerous bioactive properties due to its high phenolic compound content and is an antioxidant whose therapeutic or protective effects are investigated in organ toxicity models. This project aims to investigate the therapeutic effects of Chitosan-Silver Nanoparticle (K-AgNP) loaded Oleuropein in cellular and experimental type 1 diabetes models. In an experimental STZ-induced diabetes model, the effects on pancreatic tissue and blood glucose levels will be examined in rats following the administration of K-AgNP and/or Oleuropein for 14 days. Hematoxylin-eosin and PAS staining will be performed on pancreatic tissue, and protein expressions of GSK3β, Caspase-3, Bcl-2, NFκB, PI3K, mTOR, AKT, Glut-2 will be determined by immunohistochemical methods. In blood serum, c-peptide and insulin levels will be measured by ELISA, and MDA, GSH, and SOD levels will be detected in pancreatic tissue. The unique value of the project lies in the potential use of nanoparticle-supported Oleuropein as a solution in diabetes treatment and the contribution of the obtained results to the treatment methods of this disease.
