Ficus carica L. is one of the oldest known cultivated plants belonging to the Moraceae family. It constitutes one of the largest angiosperm genera, with over 800 species of shrubs, epiphytes and trees cultivated in various subtropical regions of the world. F. carica (FC) leaves have been used throughout history for both tea and traditional healing purposes. This study investigates FC leaf methanol extract content and its proliferation and cell viability properties on Human dermal fibroblast (HDF) cell line. F. carica leaves were collected from Bursa province of Turkey, dried, powdered and extracted. Firstly, several components of the extract were identified using Gas Chromatography-Mass Spectrometry (GC-MS) and secondly, HDF cell viability and proliferative effects (WST-8 analysis) were evaluated. Fourteen chemical components in FC leaf methanol extract were identified by GC-MS. As a result of GC-MS analysis, it was determined that the FC leaf methanol extract contained α-tocopherol (28.181%) and sucrose (22.600%); other components were β-sitosterol (13.607%), squalene (6.891%), diisooctyl phthalate (4.479%), bis(2-ethylhexyl) isophthalate (4.045%), phytol (3.803%), 1,3- distearin (3.426%), ursolic acid methyl ester (3.355%), hexanedioic acid bis(2-ethylhexyl) ester (2.625%), β-amyrin (2.366%), heptacosane (2.154%), 5β-cholestan-3-one (1.989%) andömüritose (0.479%). When the methanol extract of FC leaf was applied to HDF cell line, it was observed that cell viability decreased by 12.855% at 500 µg/mL concentration. On the other hand, it had a significant effect on cell viability and proliferation at concentrations between 15.625-250 µg/mL. As a result of these findings, it was concluded that FC leaf methanol extract increased cell proliferation and viability.
