The aim of this study was to investigate the effects of parecoxib on the ischemia/reperfusion (I/R) injury induced in rats by torsion-detorsion. Parecoxib was injected to all the rats intraperitoneally once prior to the torsion-detorsion. The ischemia/reperfusion and sham surgery groups were given the same volume of distilled water. One h later, torsion and detorsion were performed, each for two h, on the ischemia/reperfusion + 10 mg/kg parecoxib, ischemia/reperfusion + 20 mg/kg parecoxib, and ischemia/reperfusion groups. The results of the biochemical test showed that 20 mg/kg dose of parecoxib was more effective than the 10 mg/kg dose in inhibiting the increase in the MDA, COX-2, IL-1 beta and TNF-alpha levels and decrease in tGSH and COX-1 in the testicular tissue treated with torsion-detorsion. Furthermore, it was also histopathologically shown that the 20 mg/kg dose better prevented torsion-detorsion injury. The experimental results suggest that parecoxib will be clinically useful in the treatment of testicular I/R injury due to torsion-detorsion.
