Preparation and in vitro characterization of a new dosage form for antibiotic delivery: Cellulose-based gelispheres

Recently, interest in floating systems for gastroretentive purposes has considerably increased. These systems float in the stomach and provide controlled and prolonged release of the active substance from the dosage form. This study aims to prepare and characterize hydrogel gelispheres using sodium carboxymethyl cellulose. Ampicillin sodium, a broad-spectrum antibiotic, was used as a model drug. Within the scope of the characterization study, drug content, buoyancy, morphology, size, swelling, and release were evaluated. In addition, instrumental evaluation was performed with FTIR, XRD, and BET analysis. All gelispheres had a spherical structure, and different drug contents were found. Additionally, all formulations floated for 24 hours. The G7 formulation, which had a very high drug content and swelling ratio, provided extended release of ampicillin sodium. No interaction was observed with FTIR analysis and XRD analysis showed that ampicillin sodium was amorphous. As a result of the BET analysis, the G7 formulation in the mesoporous structure was shown to have a very small pore diameter. Overall, the gelispheres were successfully prepared and characterized. However, in the future, its effectiveness should be investigated in more detail by conducting further in vivo studies.
